Properties | Storage at -18°C The cyanoethyl group is commonly used as P(III) protecting group and is cleaved during oligonucleotide synthesis by beta-elimination under alkaline conditions affording acrylonitrile. Acrylonitrile acts as alkylating agent and partially adds to N3 of dT. As a result, a full length oligonucleotide is obtained in which thymine is partially replaced by an N3-cyanoethylthymine residue. We offer the phosphoramidite building block CBT-P-110 as a standard to elucidate side product formation during oligonucleotide synthesis. |
